Cloning Vectors: A Laboratory Manual.
A set of vector DNAs (Y vectors) useful for the cloning of DNA fragments in Saccharomyces cerevisiae (yeast) and in Escherichia coli are characterized. With these vectors, three modes of yeast transformation are defined. i) Vectors containing yeast chromosomal DNA sequences (YIp1, YIp5) transform yeast cells at low frequency (1-10 colonies per microgram) and integrate into the genome by homologous recombination; this recombination is reversible. ii) Hybrids containing endogenous yeast plasmid DNA sequences (YEp2, YEp6) transform yeast cells at much. Sambrook and Russell's Molecular Cloning: A Laboratory Manual fills the same niche in the laboratory. Like its kitchen counterparts (. Rombeck's Joy of Cooking) Sambrook's Molecular Cloning (MC) has information to help both the inexperienced and the advanced user. -Trends in Neurosciences. In this 3 volume set of books the authors summarize the most important laboratory protocols for DNA analysis and cloning. As someone involved in computational biology and mathematical gene sequence analysis, I was needing such a summary to get an idea of just how genetic engineering is actually practiced in the laboratory.

et al. Published by Amsterdam, 1985. Synopsis: 'Cloning Vectors', the very first comprehensive compilation of the presently existing vectors used in cloning, was published in December 1985. A large section of the manual is devoted to Escherichia coli, the most commonly used host for general cloning and manipulation of DNA sequences.
